Subject: [NMusers] Multiple dosing
Date: Wed, May 18, 2005 5:31 pm
Dear NM users,
I am currently working on an oral drug that is being given
as 10 doses at an interval of 24h and the concentrations are measured
after the 10th dose. Since there are 9 prior doses to the observations
made before the 10th dose, how do I set up multiple dosing file? Also
the population has some subjects that have observations made on them
when the drug has not reached steady state, how do I take this into
account. I tried the AMT and II data items but, I can't get it
running. Can anybody please help me setting up the data file?
Thanking You
Kumar

Kumar, why don't you share a representative piece of your data file and
control file? Then people will be able to help you a lot better.
best regards, Liping

Bharath,
See if the following case study answers your question.
http://www.accp1.org/pharmacometrics/PDF/CSIINONMEMfinal.pdf
There are some additional codes available in the code library
( http://www.accp1.org/pharmacometrics/codelib.htm).
Hope it helps.

Hi, Kumar,
hope this is not a "representative" piece of your data file, for
it misses DV item, and EVID records indicate there is no observation.
Notice your DOSE and AMT are not equal. Having both in the data file
but not using DOSE in your control steam is inconsequential, because
NM will use AMT for input amount. But make sure the number under AMT
is the true amount you want NM to use.
If you list TIME like this, there should be no difference for people who
had observations only on the 10th day and people who had observations prior
to that. If you want to use II item (so the dosing record would be simplified,
and you have enough confidence that the subject is in steady state), use
it in conjunction with SS data item.
There are many reasons a code could fail to run. It is always easier if you
attach data file and control file with your question.
Best regards, Liping
