Hi,
I have a question regrading AUC calculation for a multiple oral dose for 28
days. The oral dose was administered daily for 28 days in rats. But the dose
was adjusted for animal body weight every week. So the dose has been
changing during the 28 days period. I want to calculate AUC for 28 days in
each animal. Is there any code that can be used to calculate AUC for 28
days?

Hi Sreenivasa:
You can use NONMEM to calculate the pre-dose concentration (CMIN) for each
day. Then use a do loop with the following codes to calculate the daily AUC
and cumulative AUC out site of NONMEM (I used SAS code). It is based on the
superposition theory, Cmin1 is the concentration 24-hr post a single dose.
It is based on a one-compartment model.

CMIN1= DOSE*KA/(V*(KA-KEL))*(EXP(-KEL*24) - EXP(-KA*24));
AUC24= DOSE/CL - CMIN1/KEL + CMIN*(1 -EXP(-KEL*24))/KEL;
CUMAUC=CUMAUC+AUC24;
