Hi Xiaoli,
As someone else has already mentioned, find out what causes the second
peak. Many times it is due to enterohepatic circulation, which can be
modeled rather simply (See PK and PD Data Analysis, Concepts and
Applications by Gabrielsson and Weiner, 3rd edition, PK40). Essentially
you add a bile compartment, to which the drug distributes and from which
it is excreted back to the gut compartment.

What models could I use? I tried two compartment models, but it couldn't
