CMT1= depot, CMT2=plasma, CMT3= biomarkerA, CMT4=biomarkerB, yes. so you are going to have got 1 central compartment + a depot, so as parameters: ka between the two, CL from the central, and V for the central, that s why I don't think you can write V2. the scale seemed to be on the peripheral compartment which you have removed, you can either write only S3=1 S4=1, either add S1=V. about the subroutine, advan 6 includes a peripheral compartment, advan 2 (trans 2) might be better adapted, but because of biomarkers, I don't know...
