I would also like to know the answer. I can only report what I heard and this is not first hand experience. In general, sequential PKPD modeling is more prevelant, although there are cases when one might want (or have) to use simultaneous PKPD modeling. I remember a presentation by Mats Karlsson et al, who compared these 2 approaches (1. post-hoc estimates only used to drive PD versus 2. post-hoc estimates + raw PK data driving PD). If my memory serves me right Mats obtained different results for the 2 approaches. I think they found that method#2 was better. The reasons are not obvious to me. But may be Mats can direct us to the reference and/or help us to understand better about these approaches. On the other hand, have you tried both approaches on your specific dataset?
