RE: Single timepoint PKPD
Dear Bernard -
I would suggest you start with the Users Guide provided with your nonmem
installation. For a basic models there are excellent examples covering the
theory, implementation and code examples for how to construct the control
stream.
For additional help there are a couple other resources that may provide useful
(although the Users Guide is very thorough):
* I would also suggest searching the NMusers archive - many problems
have been solved over the years with snippets of code as well as solutions
posted.
* The accp pharmacometrics site has a couple case studies in the
modeling section that include nonmem control streams. Be aware that they are
slightly dated and refer to nonmem VI - though changes for your purposes would
most likely be minor http://accp1.org/pharmacometrics/index.html
* Some modeling environments such as Pirana offer 'template' control
streams that you can use to start to get an idea of what is necessary.
Finally, if you are looking for more in-depth explanations about both theory as
well as examples including control streams, Metrum Institute offers
well-designed online independent study courses in population PKPD modeling.
Courses are split into lecture and live demo (lab sessions) in which you can
pick up a lot in 'proper' methodologies behind both constructing and
troubleshooting your model. If you have little experience in modeling I would
highly recommend the Essentials of Population PK-PD modeling and Simulation
found here: http://training.metruminstitute.org/course/category.php?id=11

Subject: [NMusers] Single timepoint PKPD

May you kindly assist? I am learning PKPD modeling. The problem is I intend to
perform covariate modeling from a single timepoint per individual PKPD data of
the Efavirenz drug using a single compartment mechanistic model. Covariates are
genotype, sex, weight and age. What are the general features for such a control
stream in NONMEM?
