How to fit Skin Data

Hello, I am working on animal data and has sparse observation of PK from skin. I want to use NLME modeling, where the dosing is PO. Please advice how I can get a fit. I did try to add another periferal compartment and add Cabs. from that, but it failed. Thanks for your time regards Rakesh --
