single observation

Dear Nonmem-user, I have a large sample size study, but only single observation for a drug concentration and single observation for its effect at steady state is available per subject. As many parameters had been fixed to prior information, model mis-specification is the majer issue i met. So, On which criteria should should I rely to evaluate the model, specially the pharmacodynamic one? Any suggestion ? many thanks khaled
