Here is the Avastin mouse-to-man PK/PD extrapolation: Efficacy and Concentration-Response of Murine MAb VEGF in Tumor-Bearing Mice and Extrapolation to Humans.? J. Mordenti, K. Thomsen, H. Chen, Y. G. Meng, V. Licko, N. Ferrara.? Toxicol. Pathol. 27:14-21, 1999.? This work was conducted in 1996 and was submitted with the Avastin IND to support Phase I dose selection/escalation. ?This preclinical study correctly predicted the marketed dose of Avastin, which was approved in 2004.?? Figures 1 and 9 tell the whole story.? ? Prediction: 2 mg/kg per week or slightly greater than double the dose every two weeks.
