When to study BOV?

If one has a model based on parameters such as Cl, Vc, Q and Vp then how to decide on which parameters to study BOV? What prior pharmacokinetic information should be factored in before estimating this parameter? Also I think the amount of information available on different occasions influences the estimation of this parameter.
