Your error structure should have a suitable switch for CMT 1 and 2. Also you can use a hypothetical volume for your metabolite or set the scaling factor same as for your drug. In case you know the bioavailability or fraction of drug converted to metabolite you can also apply the factors accordingly. I hope this helps Atul
