Abstract: Oncology drug development has traditionally relied on categorical metrics of tumor response and disease progression. Longitudinal data on tumor size and other biomarkers of disease burden can be used to develop models of disease progression with model-based estimates of drug effect. These models can be used to discover more powerful endpoints for clinical trials and predictive markers of response to therapy. The potential of these models will be discussed using examples in renal cell carcinoma, colorectal cancer, and castrate-resistant prostate cancer.
