Abstract: With the increased number of therapies for hypertension focusing
on the renin-angiotensin system (RAS), there is evidence that some therapies
are more effective at preventing end organ damage. Although the role of
angiotensin II (Ang II) is well known and the systemic concentrations well
characterized, the dynamics of Ang II within the kidney itself are less well
characterized. This talk will walk through the development of a mechanistic
model of the RAS that was built to predict the effects of different types of
therapies on the concentrations of Ang II. The focus will be on teasing out
RAS dynamics in the renal circulation, a location that is difficult to
directly assay local peptide concentrations. Alternative sources of data for
calibration and validation will be discussed, as well as the research
applications of the model.
