Dear Users I am new to NONMEM and have been reading the NONMEM manuals but am still somewhat confused. Would appreciate any help. I have concentration data following multiple dosing - so that patients are at steady state. My compartment model is 3 compartment (which includes the compartment into which drug is administered). A few questions: My data table at present looks like - I have separated it out here to make it easier to read (however it means that the columns are slightly out of alignment) ID DATE=DROP TIME AMT RATE CMT MVD EVID CON Some comments to help you know what is going on. 1 9/26/2006 7.59 750 0 1 1 1 0 ..Dose being taken at 8am and 8pm 1 9/26/2006 19.59 750 0 1 1 1 0 ..Dose administered to compartment 1 1 9/27/2006 7.3 0 0 0 0 0 1.12 ..Trough concentration 1 9/27/2006 7.59 750 0 1 1 1 0 1 9/27/2006 8.3 0 0 0 0 0 12.67 ..Half-hour post dose concn etc up to 12 hours 1 9/27/2006 9 0 0 0 0 0 9.89 1 9/27/2006 9.29 0 0 0 0 0 7.42 1 9/27/2006 10 0 0 0 0 0 3.64 1 9/27/2006 10.3 0 0 0 0 0 2.62 1 9/27/2006 10.59 0 0 0 0 0 1.95 1 9/27/2006 12 0 0 0 0 0 0.98 1 9/27/2006 13 0 0 0 0 0 0.62 1 9/27/2006 13.59 0 0 0 0 0 0.64 1 9/27/2006 15 0 0 0 0 0 0.64 1 9/27/2006 16 0 0 0 0 0 0.59 1 9/27/2006 16.59 0 0 0 0 0 1.28 1 9/27/2006 18 0 0 0 0 0 1.62 1 9/27/2006 19 0 0 0 0 0 2.62 1 9/27/2006 19.59 0 0 0 0 0 3.03 Question 1: Do I need a column SS and if so what should I input for this? I am using ADVAN6 TOL=3 and SS=6 in my subroutine. Question 2: I suspect I should not use date and time - please advise. Question 3: Am getting an error message which is telling me that I don't have dose data and the run has stopped because of this. Thanks in advance for any help. Denny
