Dear Dick, you can use Advan 3 to fit a 2-comp model. The PK model will be the first comp, and the PD model will be the second comp. Keo(for the effect comp) will be K21. Using exponentail model for inter-individual variability, you have to use FOCE method to estimate your PK/PD parameters correctly. By the way I did use Sigmoid Emax model with effect comp, using Advan 3, but each run took up to 2-3 hours, may be yours will be faster because you do not have gamma in the Emax model.
